Diese Proteine assoziieren an die Mitochondrien- und Kernmembranen und wirken möglicherweise als spezifische Kanäle und regulieren die mitochondriale Permeabilitätstransition (Antonsson et al., 1997; Van der Heiden et al., 1997). Induktion von Bax führt zur Abnahme des mitochondrialen Transmembranpotentials und zu apoptotischem Zelltod (Pastorino et al., 1998). Dabei wurde auch die Freisetzung pro-apoptotischer Faktoren z.B. Cytochrom C aus den Mitochondrien in das Zytoplasma beobachtet (Pastorino et al., 1998; Jürgensmeier et al., 1998). [...] In einem direkten Zusammenhang mit Apoptose steht auch das Tumorsuppressorgen p53. Eine p53-abhängige Transkription wird bei verschiedenen Genen beobachtet. Es wurde berichtet, dass p53 an die Promotorregion von Angiotensinogen, AT1 Rezeptor und Bax bindet (Leri et al., 1998; Kirshenbaum et al., 1997). Die Bindung von Angiotensin II an AT1 löst Apoptose aus (Kajstura et al., 1997; Leri et al., 2000). Vermutlich kann p53 den apoptotischen Prozess transkriptionsabhängig und -unabhängig aktivieren (Ding et al., 2000).
Weiterhin wird von Cytochrom C-unabhängiger Induktion von Apoptose durch p53-abhängige Erhöhung von reaktiven Sauerstoffspezies und damit Senkung des mitochondrialen Membranpotentials berichtet (Li et al., 1999). Es gibt weitere Kontrollmechanismen der apoptotischen Signalkaskade, die auf der Ebene der Caspase-Kaskade einsetzen. Es sind verschiedene Moleküle bekannt, die als Inhibitoren für Caspasen wirken. Dazu gehören unter anderem Mitglieder der IAP (Inhibitor of apoptosis) Familie, wie c-IAP-1 und c-IAP-2 (Yaoita et al., 1998; Roy et al., 1997). Auch die Aktivierung von Mitgliedern der Mitogen-aktivierten Proteinkinasen (MAPK)-Familie kann in Kardiomyozyten zu apoptotischem Zelltod führen. Zellulärer Stress kann bestimmte Mitglieder der MAPK-Familie aktivieren (Komuro et al., 1996; Bogoyevitch et al., 1996; Aoki et al., 2002). Extrazellulär regulierte Kinase (ERK), c-Jun N-terminale Kinase (JNK) und p38-MAPK werden bei Stress aktiviert (Bogoyevitch et al., 1996; Clerk et al., 1998).
