Die Wirkung von EPO wird durch die Bindung des Hormons an einen Rezeptor auf der Oberfläche der Zellen des BFU-e- und CFU-e-Zellen-Kompartiments vermittelt (Jelkmann, 1994; Samtleben & Gurland, 1990; Adamson, 1996). Durch die Bindung an den Rezeptor wird eine Zunahme der RNA II-Polymerase-Aktivität bewirkt, gefolgt von einer Steigerung der DNA- und RNA-Synthese. Die Transkription der Globin-mRNA wird induziert (Samtleben & Gurland, 1990). Der EPO-Effekt im Knochenmark auf die EPO-abhängigen Zellen wird durch Prostaglandine vom Typ E verstärkt (Fisher et al. 1980). Vermutlich wird durch die Bindung von EPO ein Anstieg des intrazellulären Kalziums induziert (Miller et al. 1990; Yelamarty et al. 1990) und die Tyrosinkinase C aktiviert (Mason-Garcia et al. 1990).

Durch die Bindung an den Rezeptor wird eine Zunahme der RNA II-Polymerase Aktivität bewirkt, gefolgt von einer Steigerung der DNA- und RNA-Synthese. Die Transskription der Globin-m-RNA wird induziert (183). Der Erythropoietin-Effekt im Knochenmark auf die erythropoietinabhängigen Zellen wird durch Prostaglandine vom Typ E verstärkt (57). Vermutlich wird durch die Bindung von Erythropoietin ein Anstieg des intrazellulären Kalziums induziert (146; 223) und die Thyrosinkinase C aktiviert (134).
